Neurotoxicity of tryptophan metabolites.

چکیده
The metabolism of tryptophan by the kynurenine pathway leads to the production of several neurotoxic compounds, some of which have been associated with neurological disorders. Recent investigation of some relevant compounds in this pathway has provided further evidence of their neurotoxicity.

GC–MS-based metabolomics identifies an amino acid signature of acute ischemic stroke
Ischemic stroke is a serious public health problem worldwide. Here, we characterized the metabolite features of acute ischemic stroke (AIS) using a gas chromatography-mass spectrometry-based metabolomics. Forty AIS patients and 29 sex and age-matched controls were recruited. The serum metabolic profiles were significantly different between the two groups. Compared to the controls, the patients ...
متن کاملQuinolinate-induced cortical cholinergic damage: modulation by tryptophan metabolites.
Certain products of tryptophan metabolism interact with excitatory amino acid receptors to produce or protect against excitotoxicity. In this study, the action of several tryptophan metabolites, yielded by the kynurenine pathway, on cortical cholinergic toxicity was evaluated following focal injection into the rat nucleus basalis magnocellularis (nbM). Metabolites were injected singly or in com...
متن کاملDNA Binding and its Degradation by the Neurotransmitter Serotonin and its Structural Analogues Melatonin and Tryptophan: Putative Neurotoxic Mechanism
Over the years, DNA damage has been suggested to be directly linked to mutagenesis, carcinogenesis and ageing. There is ample evidence suggesting that considerable DNA damage may be induced by metabolites produced during normal or aberrant metabolic processes. Herein, we examine the chemical basis of cytotoxicity of some endogenous metabolites. Towards this goal, we study the DNA reactive activ...
متن کاملCharacterization of the kynurenine pathway in human neurons.
The kynurenine pathway is a major route of L-tryptophan catabolism producing neuroactive metabolites implicated in neurodegeneration and immune tolerance. We characterized the kynurenine pathway in human neurons and the human SK-N-SH neuroblastoma cell line and found that the kynurenine pathway enzymes were variably expressed. Picolinic carboxylase was expressed only in primary and some adult n...
متن کاملCirculating Antibodies to IDO/THO Pathway Metabolites in Alzheimer's Disease
In Alzheimer's disease, indoleamine 2,3-dioxygenase and tryptophan hydroxylase are known to induce an overproduction of neurotoxic compounds, such as quinolinic acid and 3-hydroxykynurenine from the former, and 5-hydroxytryptophol and 5-methoxytryptophol from the latter. Other compounds, such as kynurenic acid, serotonin, and melatonin are produced via the same pathways. An improved ELISA metho...
